Blood:靶向细胞因子检查点可增强脐带血CAR-NK细胞的适应性

通过CRISPR/Cas9敲除CISH可增强分泌IL-15的脐带血来源的CAR-NK细胞的代谢适应性和抗肿瘤活性。

免疫检查点疗法显著改善了某些癌症的治疗预后。细胞因子抑制剂信号家族蛋白在NK细胞的生理活性和抗肿瘤毒性中具有重要作用。CISH基因编码的细胞因子诱导性包含SH2结构域的蛋白(CIS)是该家族成员。

为了扩展检查点靶向的临床效果,Daher等设计了一种联合策略,即将白细胞介素(IL)-15-CAR工程与敲除CISH的脐带血(CB)来源的NK细胞结合起来。这一联合策略可通过增强Akt/mTORC1轴和c-MYC信号来促进NK细胞效应器功能,从而增加有氧糖酵解。

淋巴瘤小鼠模型中测试时,这种联合方法较单用其中一项更能提高NK细胞的抗肿瘤活性,在没有任何可检测的毒性迹象的情况下根除掉了淋巴瘤异种移植。

因此,研究人员得出结论,靶向细胞因子检查点可通过促进其代谢适合性和抗肿瘤活性进一步增强分泌IL-15的CAR-NK细胞的抗肿瘤活性。这种结合的方法代表了开发用于癌症免疫治疗的新一代NK细胞的一个有希望的里程碑。
