NAT COMMUN:老药新用,同时抑制肿瘤线粒体和血管生成

至今为止,癌症仍然是世界范围内的一个重要的健康问题。尽管已经存在结肠癌和乳腺癌的标准协议和疗法,但是不能满足治疗需求,建立新的治疗方法是当务之急,这可以尽量减少癌症造成的社会和经济负担。

增强的有氧糖酵解是癌症的标志之一,糖酵解本身已被提出作为一个潜在的化学治疗目标,以治疗癌症。

线粒体代谢是针对肿瘤生长机制的一个有希望的靶点。在此,研究人员对FDA批准的库进行了筛选,以确定抑制线粒体呼吸的药物。β1阻断剂nebivolol通过协同抑制线粒体呼吸链Complex I和ATP合成酶的活性,特异性地阻断癌细胞的氧化磷酸化。

机制研究显示,药物对Complex I的抑制是通过干扰NDUFS7的磷酸化来介导的;对ATP合成酶的抑制是通过ATP酶抑制因子1(IF1)的过度表达和与该酶的结合来发挥的。

值得注意的是,nebivolol还可以通过抑制内皮细胞增殖来抑制肿瘤血管生成。

总的来说,该研究证明,靶向线粒体和血管生成可以引发代谢和氧化应激危机,限制结肠癌和乳腺癌的生长。Nebivolol在癌症患者的治疗上有很大的前景,值得进一步研究其老药新用价值。
