为了研究EC特异性的转录基础,Paik等人分析组织特异性小鼠EC的单细胞RNA测序(scRNA-seq)数据,使用了多种生物信息学工具来发现scRNA-seq数据中EC异质性的标记和来源。

分析结果发现,由scRNA-seq或批量RNA-seq产生的组织特异性EC转录组测序具有强相关性,验证了该方法的可靠性。研究人员发现,有的组织(如肝、脑)的EC具有较强的组织特异性,而有的组织(如脂肪、心脏)与其他组织的EC在转录组存在相当大的重叠。此外,研究人员还鉴定出了组织特异性EC的新标志物和可能参与维持其身份的信号通路。性别在内皮细胞转录组中是一个相当大的异质性来源;Lars2是一个在雄性小鼠EC中高度富集的基因。研究人员还发现小鼠心脏和肺EC的标志物在人胎儿心脏和肺EC中保守。最后,通过分析配体和受体表达模式,研究人员确定了组织特异性EC和其他类型细胞之间潜在的血管分泌相互作用。

总之,本研究通过scRNA-seq数据揭示了维持组织特异性EC身份的转录网络,并明确了组织特异性EC间的血管分泌和功能关系。
