NEJM:中和抗体LY-CoV555治疗门诊轻中度新冠肺炎患者效果不显著

仅2800mg剂量下,中和抗体LY-CoV555可显著减少新冠肺炎门诊患者的病毒水平,其他2个剂量的效果不显著

虽然多数严重急性呼吸系统综合征冠状病毒2型(SARS-CoV-2)导致的2019年冠状病毒病(Covid-19)患者的症状是轻微的,但部分重症患者会出现危及生命的并发症。病毒中和单克隆抗体可减少病毒载量,改善症状,防止轻症患者住院。近日研究人员考察了LY-CoV555对门诊轻中度新冠肺炎患者的疗效。
 
II期临床研究中,452名新近诊断为轻度或中度Covid-19的门诊患者参与,接受单次静脉注射中和抗体LY-CoV555(700 mg,2800mg,或7000mg)或安慰剂,并评估定量病毒学终点和临床结果。研究的主要结果是第11天病毒载量与基线相比的变化。
 
在中期分析时,观察到的整个人群对数病毒载量比基线水平下降了3.81,病毒RNA消除率在99.97%以上。对于接受2800mg剂量的LY-CoV555的患者,与安慰剂相比,较基线水平下降了-0.53,病毒载量降低了3.4倍,但与安慰剂相比,700mg剂量组(-0.20)或7000mg剂量(0.09)的变化差异较小。在第2天到第6天,接受LY-CoV555治疗患者的症状严重程度略低于安慰剂组的患者。在LY-CoV555组和安慰剂组中,与Covid-19相关的住院或急诊率分别为1.6%和6.3%。
 
II期临床研究发现,仅2800mg剂量下,中和抗体LY-CoV555可显著减少新冠肺炎门诊患者的病毒水平,其他2个剂量的效果不显著。
