为了揭示动脉粥样硬化过程中SMC转分化的轨迹,并确定相关的治疗分子靶标,研究人员将小鼠和人动脉粥样硬化斑块的SMC命运图谱与单细胞RNA测序联合起来。此外,研究人员还在体外SMC衍生细胞中进行了细胞生物学实验,整合了人类基因组学研究,并进行了药理研究。

单细胞测序发现动粥中的多种SMC来源细胞类型,包括SEM细胞

结果发现,在动脉粥样硬化过程中,SMC会先转变为中间细胞状态,在人的颈动脉和冠状动脉粥样硬化斑块中也发现了这一现象。SMC衍生的中间细胞,被称为“ SEM”细胞(干细胞,内皮细胞,单核细胞)具有多能性,可以分化为巨噬细胞样和纤维软骨细胞样细胞,并可返回SMC表型。维甲酸(RA)信号被鉴定为SMC到SEM细胞过渡的调节剂,并且RA信号在有症状的动脉粥样硬化患者中失调。

SEM细胞表现出复杂的分子和细胞特征

人类基因组学发现,对冠状动脉疾病的信号进行全基因组关联研究,信号在RA信号靶基因位点上富集,并与冠状动脉疾病风险等位基因和这些基因表达受阻相关。

激活RA信号可抑制SEM细胞标志物表达、抑制SMC向SEM转化和动脉粥样硬化

用全反式RA(一种用于急性早幼粒细胞白血病的抗癌药)激活RA信号传导,可阻止SMC转化成SEM细胞,减轻动脉粥样硬化负担,并促进纤维帽的稳定性。

总之,该研究显示,细胞特异性命运、单细胞基因组学和人类遗传学的整合为SMC生物学的复杂性增添了新见解,揭示了动脉粥样硬化性血管疾病中SMC转化的靶向治疗的调控通路。
