Nat Commun :肝细胞核因子4A调节含硫氨基酸代谢并决定肝癌对甲硫氨酸限制的敏感性

含硫氨基酸代谢在肝癌中失调意味着甲硫氨酸限制有可能成为潜在肝癌治疗手段,但目前对这种失调的分子机制并不了解,甲硫氨酸限制是否可以用于肝癌治疗或增加现有疗法的敏感性值得探索。

作为一种含硫氨基酸,甲硫氨酸是人体必需氨基酸,只能从食物特别是动物蛋白中获取。已有研究表明,限制甲硫氨酸不仅在动物中有延长寿命、降脂和增强胰岛素敏感性的功效,还可能对多种癌症有抑制作用。但是,不同的癌症细胞对甲硫氨酸有不同依赖性,对甲硫氨酸限制也有不同反应,其中的分子机制尚不清楚。

肝脏是代谢含硫氨基酸的主要器官,负责代谢50%以上食源甲硫氨酸。通过肝腺苷转移酶1A(MAT1A),进入肝脏的甲硫氨酸被转化成S-腺苷甲硫氨酸,再经过转甲基和水解反应形成同型半胱氨酸,接着三甲基甘氨酸-同型半胱氨酸S-甲基转移酶 (BHMT)催化其甲基化返还为甲硫氨酸。同型半胱氨酸还可进入转硫通路,通过胱硫醚β合成酶(CBS)和胱硫醚γ裂解酶 (CTH)形成胱硫醚、半胱氨酸和硫化氢。半胱氨酸还可用于产生还原型谷胱甘肽,或通过半胱氨酸双加氧酶(CDO1)形成牛磺酸。因此,甲硫氨酸不仅用于蛋白质合成,而且在调控含硫氨基酸代谢、维持氧化还原稳态以及信号传导等方面也发挥重要作用。

早期研究发现,许多与含硫氨基酸相关的代谢酶在肝肿瘤中表达下降,并且这种下降与肿瘤的恶性程度以及不良预后都有关系。含硫氨基酸代谢在肝癌中失调意味着甲硫氨酸限制有可能成为潜在肝癌治疗手段,但目前对这种失调的分子机制并不了解,甲硫氨酸限制是否可以用于肝癌治疗或增加现有疗法的敏感性值得探索。

2020年8月7日,美国国立卫生研究院环境健康科学研究所 (NIEHS) 李小玲(Xiaoling Li)研究组,联合该所生物统计和计算生物学的李乐平(Leping Li)研究组和质谱平台主任Jason Williams,以及杜克大学Jason Locasale教授和上海交通大学医学院童雪梅教授等,在Nature Communication杂志上发表题为HNF4α regulates sulfur amino acid metabolism and confers sensitivity to methionine restriction in liver cancer的研究论文。该研究通过一系列生物信息、分子和代谢分析以及细胞和小鼠实验,揭示含硫氨基酸代谢和肝细胞核因子4A(HNF4α)之间的关系。HNF4α是肝脏的一个重要转录因子,调节多种肝脏基因表达,并对肝细胞和肝功能特性有决定作用。HNF4α表达在肝癌中下降,被普遍认为是一个肿瘤抑制基因,并可能抑制上皮间质转化 (EMT)。此项研究表明,HNF4α调节含硫氨基酸代谢,并在肝癌对甲硫氨酸限制的敏感性中发挥决定性作用。

研究人员通过对肝癌细胞和肿瘤数据库的聚类分析,证实在肝癌中,从表达水平来看,含硫氨基酸代谢酶包括MAT1A、 BHMT、CBS、CTH 和CDO1,与HNF4α呈正相关,而与间质细胞标志蛋白呈负相关。染色质免疫沉淀和荧光素酶报告实验显示,HNF4α调控含硫氨基酸代谢酶转录。与富含HNF4α的上皮肝癌细胞相比,间质肝癌细胞几乎不表达HNF4α,细胞内具有不同的含硫氨基酸代谢产物,并对甲硫氨酸限制以及Sorofenib引起的细胞死亡显示抗性。敲除上皮肝癌细胞内的HNF4α直接导致含硫氨基酸代谢酶降低,细胞内含硫氨基酸代谢变得与间质肝癌细胞相似,特别是与未敲除细胞相比,转硫通路产物包括胱硫醚、牛磺酸和硫化氢的含量显着降低。并且,上皮标志基因表达下降,间质标志基因表达上升,细胞迁移性增加, 并对甲硫氨酸限制和Sorofenib引起的细胞死亡抗性增加。小鼠肿瘤移植实验也显示,低甲硫氨酸食物可抑制上皮肝癌细胞肿瘤生长,而对敲除HNF4α的上皮肝癌细胞肿瘤作用甚微。此外,敲除上皮肝癌细胞内的单个含硫氨基酸代谢酶,在EMT、细胞迁移及抗性方面,与敲除HNF4α作用相似。反之,从基因和代谢方面恢复转硫通路显着缓解HNF4α不足造成的影响。

此项研究表明,从饮食上限制甲硫氨酸摄入,不管是单独使用还是与Sorofenib联用,都可能对肝癌产生治疗效果。病人肿瘤内的HNF4α表达水平可作为生物学指标预测对甲硫氨酸限制的敏感性以及筛选临床实验候选人。
