Curr Biol:科学家揭示调控和促进觉醒的神经机制

饥饿能够引起睡眠觉醒状态的改变,这一过程可能与能量平衡稳态、摄食行为、觉醒系统、情绪调节、奖赏系统等多个功能系统密切相关。丘脑室旁核(PVT)是重要的丘脑核团,参与了睡眠觉醒、摄食和奖赏等日常行为的调节。

论文第一作者、华中科技大学博士化瑞芳告诉《中国科学报》记者,以往电生理研究发现PVT神经元电活动存在昼夜差异。PVT与视交叉上核的双向联系也提示其在生物节律调节中的作用。由此可推测PVT脑区可能在协调睡眠和摄食行为中起着重要的整合作用。

为阐明PVT脑区在上述行为调节中的具体神经机制,该团队采用在体多通道和多导睡眠记录技术,结合光遗传学,在小鼠身上实施测验。通过观察PVT脑区神经元在饥饿状态下电活动变化,科学家发现饥饿能够提高小鼠觉醒水平,并激活PVT脑区中钙视网膜蛋白阳性神经元CR+,并且能够驱动其下游关联脑区抑制小鼠睡眠。

该研究首次揭示了PVT中存在一类重要的神经元可对饥饿产生响应,同时在协调睡眠和觅食冲突中发挥关键作用。

该团队未来计划在该脑区开展与代谢疾病相关的机制研究。通过与临床相结合,对该脑区在机体能量代谢中发挥的重要作用进行深入探讨。
