雌二醇是女性主要的性激素之一,对男女个体的骨骼健康都起着重要的作用。几种酶参与了雌二醇代谢,但很少进行全基因组关联研究(GWAS)来归纳导致雌激素水平变化的遗传促进因子。


 
近日,内分泌和代谢性疾病领域权威杂志Journal of Clinical Endocrinology & Metabolism上发表了一篇研究文章,该研究旨在明确影响雌二醇水平的基因位点,并估计雌二醇对骨密度(BMD)因果关系的影响。


研究人员对来自英国生物银行(UKB)的男性(N=147690)和女性(N=163985)的雌二醇进行了GWAS。雌二醇被分为二元表型;高于/低于检测阈值(175pmol/L)。研究人员使用孟德尔随机化进一步估计了雌二醇对BMD因果关系的影响。


研究人员确定了14个与男性雌二醇水平相关的独立位点(P<5x10-8),其中一个(CYP3A7)是全基因组的;在女性中,研究人员确定了7个名义上(P<0.05)具有显著相关性的位点。此外,研究人员还确定了一个雌性特定基因座。大多数基因座包含功能相关的基因,这些基因在以前的GWAS中尚未讨论过与雌二醇水平之间的关系。例如,SRD5A2编码参与处理雄激素的类固醇5-α还原酶,以及编码可能参与雌二醇消除的酶的UGT3A1和UGT2B7。在ABO基因座标记O血型的等位基因与较高的雌二醇水平相关。研究人员确定了高雌二醇水平对男性(P=1.58x10-11)和女性(P=7.48x10-6)BMD增加的因果效应。
