三阴性乳腺癌(TNBC)是一种最具侵略性的乳腺癌形式,其占所有确诊乳腺癌病例的15–20%,具有高转移率和高复发率。

葡萄糖代谢及其重编程是癌细胞的一个关键特征。癌细胞所表现出的增殖的增加伴随着对能量和合成代谢底物的需求增加。许多癌症通过被称为Warburg效应的新陈代谢转变来适应这些需求,其中氧化磷酸化作用受到了抑制。这种代谢的改变也与患者不良的预后和治疗抗性的产生相关。

SU212诱导TNBC细胞的有丝分裂期阻滞和细胞凋亡作用

为了规避Warburg效应,针对不同癌症的多项临床试验都采用了组合干预方法,即使用代谢重编程和包括二甲双胍在内的化学治疗剂。这些代谢干预中的大多数是通过间接激活AMPK来改变细胞代谢,从而促进有氧糖酵解的氧化磷酸化作用。因此,寻找能够激活AMPK的小分子化合物或可成为三阴性乳腺癌(TNBC)治疗的一种潜在策略。

SU212治疗可抑制肿瘤发生发展和转移

该研究开发了新型的AMPK激活剂,SU212,并检测其对TNBC细胞的治疗作用。研究人员发现,该激活剂能够选择性地诱导TNBC细胞中的氧化磷酸化并减少糖酵解作用,而并不会影响正常细胞中的这些通途径。SU212可通过激活AMPK来完成代谢重编程作用,该作用独立于能量应激的影响且与血糖/胰岛素状态无关。该作用会最终导致TNBC细胞中有丝分裂的停滞和细胞凋亡的发生。体内实验显示,SU212能够抑制肿瘤的发生发展和转移。


综上,该研究结果显示,SU212能够直接激活TNBC细胞中的AMPK,且不影响正常细胞中的葡萄糖代谢。该研究也为进一步开发用于治疗TNBC的SU212提供了令人信服的临床前数据。
