透明细胞肾细胞肿瘤(ccRCC)是最常见肾癌。因ccRCC对化疗和放疗具有较强的抗性,其预后通常较差。许多研究表明癌症干细胞/肿瘤起始细胞(CSCs/TICs)与肿瘤的发展、疾病的恶化和入侵、转移以及药物抗性有关。然而,从已建立的RCC细胞系基础ccRCC研究模型中分离出的CSCs/TICs的体内致瘤潜力尚未调查。

最近,有研究人员从Caki-1 RCC细胞系中分离出了CD105+、CD105-、CD44+和CD44- 以及CD44-/CD105- CD44+/CD105+和CD44-/CD105+细胞,证实了在稳定的细胞系中与干细胞表型相关多个亚群的存在。研究人员随后将分选的细胞皮下注射到NOD SCID小鼠中,并使用MRI和PET/CT对肿瘤生长进行监测。研究发现,CD105+、CD44+、CD44?、CD44-/CD105+和CD44-/CD105-(不包括CD105-或者CD44+/CD105+)植入后均观察到了肿瘤的生长。CD44-/CD105-细胞植入能够诱导肿瘤,并且与其他肿瘤相比具有更长的T1和不同的代谢模式。另外,所有的肿瘤均具有低的[18F]FDG摄取能力。CD105+和CD44-能够表达Nanog和Oct-4,而CD44-肿瘤还能额外表达内皮细胞标记CD31。
