T细胞受体/CD3(TCR/CD3)复合物的完整性对胸腺中T细胞的阳性和阴性筛选至关重要,同时对外周T淋巴细胞的效应和调节作用也很重要。人类CD3D、CD3E和CD3Z基因缺陷可导致严重的免疫缺陷,发病年龄早,感染风险增加。与此相反的是,CD3G突变可导致更温和一些的表型,主要的特点是自身免疫病。

但是,CD3γ在建立和维持免疫耐受中的作用尚未阐述清楚。近日Blood杂志上发表一篇文献,研究人员对携带与自身免疫病相关的CD3G遗传缺陷的患者的T细胞的异常功能进行相关研究。

利用高通量测序(HTS)对6位携带CD3G突变的患者和健康对照的Treg、传统CD4+(Tconv)和CD8+T细胞上的T细胞受体β(TRB)的组成和多样性进行研究。通过研究Treg抑制Tconv细胞增殖的能力来评估Treg的功能。CD3G突变患者的Treg细胞的多样性减少、单一性克隆增加,而且抑制Tconv细胞增殖的功能减弱。CD3G缺陷患者的Tconv细胞的TRB库,在CDR3(自我反应的生物标志物)的第6位和7位上富集疏水性氨基酸。

总而言之,本研究数据证明了携带CD3G突变的患者的T细胞库的分子特征,可能会增加与此条件相关的自身免疫病的发病率。
