AJRCCM:m6A在肺动脉高压中的作用

肺动脉高压 (PH) 是一种进行性肺血管疾病,其特征是不可逆的肺血管重构 (PVR) 和肺动脉平滑肌细胞 (PASMC) 功能障碍 。

研究背景:

N6-甲基腺苷(m6A)是mRNA上最普遍的内部修饰参与细胞过程。METTL3是用于m6A合成的编写器复合体的核心子单元,而其他子单元则对其活动和特异性有所贡献。ALKBH5和FTO是脱甲基酶,可消除mRNA的m6A甲基化,因此在动态调节m6A修饰中与甲基转移酶相对应。m6A对mRNA翻译,输出和稳定性的下游影响主要由识别修饰靶标的特定读写区确定。通过对m6A修饰转录本的翻译控制,包括癌细胞的减数分裂,细胞增殖和上皮-间质转化(EMT),已在各种生理和病理学过程中报道了YTHDF1作为m6A修饰的识别者。尽管最近m6A修饰的circRNA与大鼠低氧诱导的PH相关,但m6A修饰的临床意义和分子机制以及YTHDF1在肺动脉高压中的作用尚待研究。

研究目的:m6A修饰和m6A效应在PH发病机制中的作用。

研究方法:

在这项研究中,通过检测PAH患者和实验动物模型标本中m6A修饰YTHDF1的表达情况,将患者和动物模型的肺动脉原代平滑肌细胞进行功能分析等。我们揭示了 m6A 水平的显着增加以及 YTHDF1 蛋白在人肺动脉高压 (PAH) 和实验 PH 模型中的表达增加。 YTHDF1 抑制在体外减弱了缺氧 PASMC 的增殖、迁移和表型转换,并在体内防止 PVR 和改善右心功能。 此外,我们发现 YTHDF1 以 m6A 依赖性方式促进 MAGED1 翻译,并且 MAGED1 的基因消融改善了小鼠的动脉肌肉化、管腔闭塞和心室重构。 本文中的这些数据首次证明了 m6A 修饰的mRNA 在 PH 中的病理作用,并表明靶向 YTHDF1 或 MAGED1 的策略可能是治疗 PH 的有效疗法。

研究结果:

研究首次证明了在肺动脉高压的小鼠和大鼠模型以及IPAH患者中总RNA的m6A水平升高。在HPASMC和新型YTHDF1基因敲除小鼠中通过基因沉默靶向YTHDF1,至少部分地通过降低MAGED1的翻译来减弱肺动脉高压中的细胞增殖和血管重塑。MAGED1被鉴定为m6A调控的基因,MAGED1的基因消融改善了肺动脉高压中的血管重塑和血液动力学。m6A修饰的RNA是肺动脉高压的新型介体,靶向此途径可能是改善肺动脉高压发展的有效治疗方法。
