J Periodontol:环孢菌素A对人牙周膜干细胞的作用

本研究旨在研究环孢素A(CsA)对人牙周膜干细胞(hPDLSCs)的成骨分化,破骨细胞分化和血管生成潜力的影响。

本研究旨在研究环孢素A(CsA)对人牙周膜干细胞(hPDLSCs)的成骨分化,破骨细胞分化和血管生成潜力的影响。

收集正畸患者拔出的牙齿用于分离hPDLSC。使用3-(4,5-二甲基噻唑-2-基)-2-,5-二苯基四氮唑溴化物评估细胞增殖,碱性磷酸酶(ALP)和茜素红(ARS)染色评估成骨分化。实时聚合酶链反应(PCR)用于定量转录本。用含CsA的hPDLSC培养基处理细胞后,进行了骨髓衍生巨噬细胞(BMM)的抗酒石酸酸性磷酸酶染色和人脐静脉内皮细胞(HUVEC)的血管生成试验。使用蛋白质印迹法研究了介导血管生成活性的信号通路。

结果发现,CsA抑制了hPDLSCs的增殖,但通过ALP和ARS染色以及成骨转录本的PCR测定,增强了成骨分化。破骨细胞转录本在hPDLSCs中的表达以及含CsA组处理的BMM的分化不受CsA的影响。但是,血管生成转录本和已知支持血管生成的转录本(细胞外信号P调节激酶(ERK)和p38和c-fos的磷酸化)的表达被抑制。CsA组抑制了HUVEC的血管生成能力。

因此,CsA通过阻断hPDLSCs中的ERK和p38 / c-fos通路来增强成骨分化并减少血管生成。
