AK002是靶向Siglec-8的IgG1型单抗，通过抗体介导的细胞毒和诱导细胞凋亡，消耗嗜酸性粒细胞。

生物技术公司Allakos宣布其靶向Siglec-8的单抗lirentelimab（AK002）在嗜酸性胃炎和嗜酸性十二指肠炎（ENIGMA）患者中进行的多中心、随机、双盲、安慰剂对照的II期临床研究结果发表在新英格兰医学杂志（NEJM）上，文章题目为Anti-Siglec-8 Antibody for Eosinophilic Gastritis and Duodenitis。

结果显示与安慰剂相比，AK002的所有剂量组在预先指定的主要和次要终点（包括胃肠道组织嗜酸性粒细胞计数和患者报告的疾病症状）上均显示出具有临床意义和统计学意义的改善。

Siglec-8是选择性高表达在成熟的嗜酸性粒细胞和肥大细胞上的抑制性受体。嗜酸性粒细胞和肥大细胞的过度激活已被证实是引发嗜酸性胃炎和嗜酸性十二指肠炎的关键驱动因素。AK002作为靶向Siglec-8的IgG1型单抗，可以通过抗体介导的细胞毒和诱导细胞凋亡，消耗嗜酸性粒细胞。

嗜酸性胃炎和嗜酸性十二指肠炎是严重的炎症性疾病，其特征是在胃或十二指肠中分别存在高水平的嗜酸性粒细胞。与其他胃肠道疾病相比，这些疾病可能被严重误诊。没有专门的治疗方法，全身性类固醇激素治疗可改善症状，但由于副作用大，一般无法长期使用类固醇激素治疗。
