Nat Commun:必需omega-3脂肪酸对发育中的大脑有调节作用

发达国家和发展中国家饮食习惯的西化,伴随着饮食中n-3 PUFAs摄入量的逐渐减少。母体中n-3 PUFAs的低摄入量与人类的神经发育疾病有关。

脂类是中枢神经系统(CNS)的主要成分之一。其中,花生四烯酸(AA,20:4n-6)和二十二碳六烯酸(DHA,22:6n-3)是灰质长链(LC)多不饱和脂肪酸ω-6和ω-3(称为n-6或n-3 PUFAs)的主要形式。这两种LC PUFAs被发现主要是以磷脂的形式存在,并构成了脑细胞膜的构件。

AA和DHA是从各自的饮食前体生物合成的,包括亚油酸(LA,18:2n-6),和α-亚麻酸(ALA,18:3n-3),或直接从饮食中获得(主要是肉类和乳制品的AA,肥鱼的DHA)。

Omega-3脂肪酸(n-3 PUFAs)是大脑功能成熟的必要条件。发达国家和发展中国家饮食习惯的西化,伴随着饮食中n-3 PUFAs摄入量的逐渐减少。母体中n-3 PUFAs的低摄入量与人类的神经发育疾病有关。然而,n-3 PUFAs缺乏影响中枢神经系统发育的机制还不太清楚。

小胶质对突触的主动吞噬调节大脑发育。突触修剪受损与一些神经发育障碍有关。最近,研究人员在Nature Communications杂志发文,确定了母体n-3 PUFA摄入量低对小鼠海马发育的不利影响的分子机制。

该研究结果表明,母体饮食中n-3 PUFA的缺乏会增加啮齿动物发育中的海马突触元素的小胶质细胞介导的吞噬作用,其部分是通过激活12/15-脂氧合酶(LOX)/12-HETE信号,改变神经元形态,影响后代的认知能力。

这些发现为母体n-3 PUFAs膳食缺乏引起的神经发育缺陷提供了机制性的启示。
