J Nat Prod：重大突破！藤蔓复合物竟可杀死癌细胞！

说起胰腺癌，不得不说起一个人类科技的推动者—乔布斯。乔布斯的去世令世界哗然，更是让世人对胰腺癌感到胆颤。目前胰腺癌的诊断率尚可，但其进展速度之快令大多数医生无计可施，所以有人将其比喻为最新恐怖的“肿瘤之王”。为了治疗这一恐怖的“肿瘤之王”，各国科学家也绞尽脑汁，希望通过各种方法延缓胰腺癌患者的寿命，但效果甚微。目前来自日本Suresh Awale教授联合德国Gerhard Bringmann教授一同研究了一种来自刚果盆地的藤类植物，他们发现这种植物中含有抗胰腺癌的有效成分，其最新的研究成果发表于《Journal of Natural Products》杂志。

众所周知，胰腺癌难以治疗，并且病情前景不佳。据美国癌症协会（ACS）称，早期胰腺癌患者的5年生存率为12-14％，这在所有肿瘤类型中也属于致死率很高的类型。为此来自德国维尔茨堡Julius-Maximilians-Universit？t大学的有机化学教授Gerhard Bringmann与日本Suresh Awale教授展开了一系列研究，他们为胰腺癌的高致死率提出新的解释，并提供了一个全新的治疗方法。

Awale及其同事表示，胰腺癌抵抗治疗的部分原因是由于胰腺癌细胞即使在最恶劣的环境中也能存活。详细来说，人胰腺细胞具有在肿瘤微环境内血管处于缺氧条件下大量增殖的能力，其对于营养缺乏有显着的耐受性。换句话说，胰腺癌细胞能够吸收周围的所有能够被他们利用的氧气和营养物质，进而维持其大量扩增。

在这种“断水断粮”的严峻条件下，大多数细胞会死亡。然而，胰腺癌细胞通过激活称为Akt / mTOR的分子通路来克服这些困难，进而维持其生存。Akt / mTOR是一种细胞信号传导途径，这种信号通路可通过诱导自噬等多个机制维持细胞的正常代谢。

Awale解释说，发现能够在应激条件下抗肿瘤细胞的药物，即抗紧缩性，是研究抗癌药物新策略，旨在寻找能够在严峻条件下靶向并阻止癌细胞繁殖的化合物。Awale及其团队测试了一种名为ancistrolikokine E3的物质，它来自刚果雨林中的一种叫做Ancistrocladus likoko的藤蔓的提取物。科学家们先前的发现，刚果热带雨林中的葡萄藤所含的某些生物碱具有抗紧缩性。



在当前这项研究中，研究人员从刚果藤本植物的枝条中分离出生物碱ancistrolikokine E3并测试其对癌细胞的影响。他们发现，ancistrolikokine E3引起癌细胞形态的“显着改变”，并最终导致其大量死亡。该化合物通过抑制Akt / mTOR和自噬途径杀死癌细胞，否则这些信号通路将使癌细胞在严峻的肿瘤微环境中得以存活。此外，葡萄藤衍生化合物阻止胰腺癌细胞迁移和形成转移灶。这表明在某些剂量下，该化合物可以阻止转移的发生。



研究人员表示，这项新研究提供了某些生物碱对营养缺乏条件下的胰腺癌细胞具有抑制作用的影响的第一个实验证据。他们总结说：“Ancistrolikokine E3和相关的生物碱为在抗紧缩性条件研发新型抗癌药物提供了先导作用”。
