J Periodontal Res:抗坏血酸磷酸酯钠对成骨细胞活力和分化的影响

这篇研究的目的是为了评估SAP对成骨细胞活性和分化的作用。

抗坏血酸磷酸酯钠(SAP)是一种亲水并稳定的左旋维他命C衍生物,通过细胞磷酸酶可将其转换为游离抗坏血酸(AA),以促使它在介质中持续释放。AA参与了牙周组织的维护与愈合。它对成骨细胞的活性具有调控作用,可诱导成骨分化表型基因表达并促进细胞外基质胶原沉积。它也可以消除牙周疾病中防御细胞产生的大量活性氧。因此,这篇研究的目的是为了评估SAP对成骨细胞活性和分化的作用。

研究使用小鼠前体成骨细胞MC3T3-E1系作为实验对象。通过trypan blue dye exclusion法测定细胞活力,通过定量PCR检测成骨分化相关基因的表达。ELISA评估I型胶原的分泌,茜素红S染色检测矿化基质的形成情况。

结果显示,50至500 µmol/L浓度的SAP并未影响成骨细胞的活性,但是会刺激它们分化,诱导RUNX2, COL1A1和BGLAP2表达;I型胶原分泌水平更高;并且,与未暴露于SAP相比,施以200或400 µmol/L SAP刺激的细胞其细胞外基质的矿化明显增加。

结论:基于它的稳定和刺激成骨前体细胞分化的能力,表明将SAP整合入局部释放系统,膜/支架或生物材料,有利于骨组织的形成,进而促进牙周组织愈合。
