感染HIV-1后,每年从试验HVTN 073(C亚型,DNA/MVA,1期试验,n = 1)、HVTN 086(C亚型,DNA/MVA/gp140蛋白,1期试验,n = 2)和HVTN 204(多亚型,DNA/腺病毒血清型5 [Ad5],2期试验,n = 7)的参与者和4名安慰剂接受者中收集血清样本。分别使用酶联免疫吸附试验和TZM-bl细胞中和试验,在HIV感染前和感染后测定Env蛋白和多肽的结合和中和抗体反应。感染HIV的南非人作为未接种的对照组。

 

结果显示,从HIV-1亚型C感染的第一年开始,已检测到与gp41、V3、V2、膜近端外部区域(MPER)和CD4结合位点的结合抗体,疫苗接种者和安慰剂接受者的水平相似。在所有参与者中都检测到了针对1A级病毒的中和抗体反应,滴度最高的是C亚型病毒,MW965.26。在感染前没有观察到任何反应,表明疫苗引发的HIV特异性抗体已经减弱。在HIV感染2~3年后,观察到对第2级分离物的零星中和活性,但这些反应在疫苗组和安慰剂组以及未接种疫苗的对照组中是相似的。

综上所述,这些数据表明,事先接种这些免疫原并没有改变对HIV-1感染的抗体反应,也没有加速HIV中和广度的发展。
