Nature:母体微生物组可调控胎儿神经发育

通过研究耗尽和选择性重建的母体肠道微生物组对小鼠胎儿神经发育的影响,探究母体肠道微生物组在稳态过程中调节早期胚胎大脑发育和后代晚期行为的作用。

微生物组是大脑功能和行为的一个重要的调节器。已有的研究显示,与普通的无特定病原体(SPF)的动物相比,没有微生物菌落(无菌,GF)的动物或耗尽微生物组(抗生素处理,ABX)的动物会表现出神经生理和行为的改变。

其中,只有一个子集的表型可以通过产后恢复微生物组来纠正,这表明母体妊娠微生物组参与调节子代成年后脑功能和行为的发育过程。事实上,肠道微生物组是应对母体挑战必须的,如免疫激活,高脂肪饮食和心理社会压力,诱导小鼠的神经行为异常。

然而,至今,我们仍然不清楚,这种微生物对神经发育的影响是否起源于产前,通过破坏母体微生物群的功能,和/或产后,通过垂直传播的新生儿微生物群的改变。

此外,虽然已有的研究证明,母体微生物组可以调节宿主对急性应激的反应,但我们仍然不清楚,在没有环境挑战时,它是否会影响后代的发育。

最近,研究人员在Nature杂志发文,其通过研究耗尽和选择性重建的母体肠道微生物组对小鼠胎儿神经发育的影响,探究母体肠道微生物组在稳态过程中调节早期胚胎大脑发育和后代晚期行为的作用。

研究人员发现,来自抗生素处理的母鼠和无菌母鼠的胚胎表现出与轴突发生相关的基因的脑表达减少,丘脑皮质轴突缺失,丘脑轴突在细胞外因子的作用下生长受损等。

用特定的菌群对微生物组缺失的母鼠进行定植,可防止胎儿脑基因表达和丘脑皮质轴突发生的异常。

代谢组剖析显示,母体微生物组调控着母体血清和胎儿大脑中的许多小分子。特定的微生物群依赖性代谢物可以促进胎儿丘脑外植体轴突的生长。此外,母体补充这些代谢物可消除胎儿丘脑轴突的缺陷。

在怀孕期间操纵母体微生物组和微生物代谢产物,产生的成年后代在两个厌恶性体感行为任务中的触觉敏感性改变,但在许多其他感觉运动行为中没有明显的差异。

因此,该研究结果表明,母体肠道微生物组促进了胎儿丘脑皮质轴突的发生,可能是通过微生物调控的代谢物向大脑发育中的神经元发出信号。
