Int J Biol Sci:上调HIF-1α应对缺氧环境中骨缺损的修复

老年人骨缺损的修复仍然是现代医学面临的一大挑战。移植骨髓间充质干细胞(BMSCs)联合或不联合生物材料已成为一种有前途的骨修复和再生方法。通常,移植的BMSCs在含氧量正常的条件下(21%O2和10%血清培养基)在体外培养。然而,骨缺损区域的微环境较差,其中存在较低的生理氧张力(<1%)和组织缺血。因此,如何提高移植的BMSCs在体内低氧和缺血区的存活率和成骨至关重要。 缺氧诱导因子-

缺氧诱导因子-1α(HIF-1α)在移植后骨再生过程中对骨髓间充质干细胞的耐受性、血管生成和骨形成具有重要作用。既往研究已经证明Dimethyloxaloylglycine(DMOG)改善了BMSCs的血管生成活性。

通常,血管生成和骨生成彼此耦合。我们发现1%O2缺氧预处理和0.5mM DMOG联合处理的BMSCs显示Hif-1α的上调,可提高BMSCs在恶劣条件下(无血清培养基和1%O2)的体外存活率,并在体外1%O2微环境下增强骨髓间充质干细胞的血管新生和成骨能力。将缺氧预处理的骨髓间充质干细胞移植入老龄SD大鼠临界尺寸的下颌骨缺损,测试缺氧预处理方法的有效性。结果发现缺氧预处理的BMSCs可以改善体内临界尺寸下颌骨缺损的修复。

综上所述,这些数据表明,低氧预处理的BMSCs随着Hif-1α的上调或可增强老年人的骨愈合过程。
