持续感染人乳头瘤病毒(HPV)是大多数宫颈癌及其他几种肛门生殖器和口咽癌的主要原因。这些癌症主要是由HPV16和HPV18引起的。然而,其他13种高危HPV类型也与癌症的发生发展相关。HPV诱导的恶性肿瘤主要由病毒致癌基因E6和E7驱动,其通过调控宿主细胞信号通路,增加细胞增殖并增强细胞的存活,最终使感染的细胞发生恶性转化。

因此,了解HPV相关疾病中病毒-宿主之间的相互作用有利于鉴定新型的治疗靶标。


MAPK(促分裂素原活化蛋白激酶)能够将细胞外刺激转化为一系列的细胞反应,而ERK(MAPKs细胞外信号调节激酶)和p38目前已证实在HPV生命周期和HPV诱导的转化中具有至关重要的作用。相比之下,目前对于JNK(c-Jun N端激酶)的功能作用还知之甚少。

E6介导的JNK的激活和下游信号转导通路的相关原理图

该研究显示,在宫颈疾病和宫颈癌中JNK信号通路被激活。HPV E6癌基因通过E6 PDZ结合基序依赖性的方式诱导JNK1/2的磷酸化。研究人员发现,采用小分子抑制剂或敲低JNK底物c-Jun来阻断JNK1/2信号转导通路,可降低细胞增殖作用并诱导宫颈癌细胞的凋亡。


进一步的研究显示,该表型至少部分受JNK依赖性的EGFR信号的激活所驱动的,而该激活作用是通过提高EGFR和EGFR配体EGF和HB-EGF的表达水平来实现。JNK/c-Jun信号能够促进宫颈癌细胞的侵袭,该作用需要上皮-间充质转化(EMT)相关转录因子Slug和间充质标记物波形蛋白(Vimentin)的表达。此外,JNK/c-Jun信号是HPV E6/E7的组成性表达所必需的,而HPV E6/E7对于宫颈癌细胞的生长和存活至关重要。

综上,该研究结果显示,EGFR信号转导通路与HPV E6/E7的表达之间存在正反馈回路,该研究也确定了HPV介导EGFR信号转导并促进宫颈癌细胞增殖、存活和EMT进程的调节机制。因此,该研究或为宫颈癌治疗提供了新的潜在的治疗靶标。
