视网膜缺血是视力丧失和损伤的主要原因,并且是与青光眼、糖尿病性视网膜病和视网膜中央动脉阻塞等疾病的潜在致病机制。患者的视网膜再生能力有限。他们以前的研究表明,在视网膜缺血的大鼠模型中向玻璃体内注射间充质干细胞(MSC)和MSC条件培养基对神经具有保护作用。

MSC和条件培养基的神经保护作用可能主要由细胞外囊泡(EV)介导实现的,在体外视网膜缺血的氧 - 葡萄糖剥夺(OGD)模型中,他们测试了MSC产生的EV作用。用MSC衍生的EV处理R28视网膜细胞可显着减少细胞死亡,并且细胞增殖抑制效应得到缓解。

同时,他们在体外进行了视网膜细胞对EV内吞作用模式的机理研究。他们发现,EV内吞作用是剂量和温度依赖性的,可饱和的,并且通过由caveolar内吞途径介导的细胞表面硫酸肝素蛋白多糖起作用。在大鼠模型中,视网膜缺血后24小时,将MSC-EV注射到玻璃体液中显着增强相关功能,并减少神经炎症和细胞凋亡。EVs被视网膜神经元、视网膜神经节细胞和小胶质细胞摄取。在玻璃体内给药后,存在于玻璃体液四周,与玻璃体液组分具有饱和性的结合。
