A&R:两个独立队列中肠道微生物群与血清尿酸升高之间的关联

     目的:高尿酸血症是痛风的前兆,通常也存在于由微生物群失调促进的其他代谢疾病中。该研究探索肠道微生物群与人类高尿酸血症和血清尿酸盐水平的关系。

     方法:研究参与者来自一项以社区为基础的观察性研究——湘雅骨关节炎研究(发现队列)。高尿酸血症定义为女性血清尿酸水平>357μM/L,男性>416μM/L。使用粪便样本的16S核糖体RNA测序分析肠道微生物群。该研究检查了微生物群失调(即微生物群落的丰富度、多样性、组成和相对丰度)之间的关系并预测了导致高尿酸血症和血清尿酸盐水平的功能途径。研究者在一项独立的观察性研究Step Study(验证队列)中验证了这些关联。

     结果:发现队列由来自农村地区的1392名受试者组成(平均年龄61.3岁,57.4%为女性,17.2%患有高尿酸血症)。与尿酸正常的参与者相比,患有高尿酸血症的参与者的丰富度和多样性降低,微生物群的组成发生改变,并且粪球菌属的相对丰度较低。与尿酸正常的受试者相比,高尿酸​​血症受试者预测的KEGG代谢途径(包括氨基酸和核苷酸代谢)显著改变。肠道微生物群的丰富性、多样性和低相对丰度的粪球菌属也与高水平的血清尿酸盐有关。这些发现在480名参与者的验证队列中得到了验证。

    结论:肠道菌群失调与血清尿酸盐水平升高有关。这项研究检验了微生物群失调可能调节血清尿酸盐水平的可能性。
