BP:构建环状RNA- miRNA- mRNA网络揭示补阳还五汤治疗脑缺血的潜在机制

构建补阳还五汤治疗的大脑中动脉闭塞(MACO)大鼠的环状RNA (circRNA)-microRNA (miRNA)-信使RNA (mRNA)网络,为进一步阐明补阳还五汤治疗脑缺血的分子机制奠定基础。

背景与目的:补阳还五汤(BHD)是一种治疗脑缺血(CI)的有效中药方剂。然而,BHD在CI中的分子机制尚未完全阐明。整合BHD治疗的大脑中动脉闭塞(MACO)大鼠的环状RNA (circRNA)-microRNA (miRNA)-信使RNA (mRNA)网络。

研究方法:将SD大鼠随机分为对照组、模型组、模型+BHD组(2.5、5、10 g/kg)、模型+丁苯酞(NBP)组(54 mg/kg)。采用改良神经严重程度评分系统(mNSS)评价大鼠神经功能。Nissl染色评价病理病变,免疫组织化学评价BHD对神经血管单元(NVU)相关蛋白微管相关蛋白2 (MAP2)、胶质原纤维酸性蛋白(GFAP)和血管性血友病因子(VWF)的影响。使用CeRNA和miRNA微阵列建立环状RNA、miRNA和mRNA谱,最后构建环状RNA - miRNA - mRNA三元转录网络

 

研究结果:

  • 神经行为试验结果显示,与对照组相比,模型组大鼠神经行为评分显著升高;与模型组的大鼠相比,BHD治疗组大鼠的神经行为评分明显下降,尤其是中剂量和高剂量组,表明BHD改善了CI大鼠的神经行为评分
  • Nissl染色显示,与对照组相比,模型组皮质区和海马齿状回神经元排列不规则,细胞间隙增宽,细胞核固缩。与模型组相比,三剂量BHD组和NBP组大鼠皮质区和海马齿状回神经元排列较为整齐,细胞间隙减少,核仁清晰,表明BHD改善了缺血脑组织的病理学改变
  • 神经血管单元(NVU)由神经元、胶质细胞和血管组成,常用MAP2、GFAP和VWF分别标记成熟神经元、成熟胶质细胞和内皮细胞。免疫组化结果显示,MCAO大鼠缺血侧脑组织MAP2、VWF表达降低,GFAP表达升高,BHD治疗逆转上述变化,表明BHD保护MCAO大鼠脑缺血侧的NVU
  • 分别鉴定了7个、3个和86个显著失调的环状RNA、miRNA和mRNA,它们与BHD的神经保护作用相关。生物信息学分析表明,这些靶点可能通过多种途径发挥治疗作用,如VEGF和Hippo信号通路。最后构建了circRNA-miRNA-mRNA网络,BHD可能通过RNO_CIRCpedia_5686和RNO_CIRCpedia_3280靶向吸收rno-miR-211-3p,进而调控8个mRNA的表达,从而解释了其抗CI作用。

研究结论:BHD改善了MCAO大鼠的神经行为评分,并保护了缺血侧脑组织的NVU。研究BHD对CI大鼠环状RNA、miRNA和mRNA微阵列的影响,初步构建环状RNA-miRNA-mRNA三元转录网络。进一步分析表明,RNO_CIRCpedia_5686/miR-211和RNO_CIRCpedia_3280/miR-211在促进BHD改善CI后NVU的作用中起关键作用。该研究为进一步阐明BHD治疗CI的分子机制奠定了基础。
