Nat Commun:绿脓菌素具有治疗线粒体疾病的潜力

最近,研究人员表明,绿脓菌素,一种来自细菌的氧化还原酶,可以取代复合体III的氧化还原功能,起到电子分流的作用。

线粒体负责合成细胞功能所需的大部分ATP。ATP产能的缺陷通常对肌肉和大脑等能量需求较高的组织危害性更大。ATP合成的减少可能是由于呼吸链(RC)的组成部分,即参与氧化磷酸化(OXPHOS)的复合物I(CI)、II(CII)、III(CIII)、IV(CIV)和ATP合成酶(复合物V)的功能受损导致的。电子通过RC传输产生的能量被CI、CIII和CIV利用,将质子逆着电化学梯度泵过线粒体内膜(IMM)。这样产生的质子动力再通过质子沿电化学梯度的再进入来驱动ATP的合成,从而促进复合物V的旋转活化,质子梯度还可以进行其他相关的过程,如小阳离子的转运,而跨膜电位是蛋白质穿过IMM进入基质的运输所需要的。

线粒体疾病是指通过破坏RC或ATP合成酶的复合物,或通过影响线粒体DNA(mtDNA)的复制/转录/翻译、RC辅助因子的生物合成和线粒体生物发生而影响OXPHOS的遗传缺陷。电子流的抑制不仅导致OXPHOS的损伤和ATP合成的减少,而且还会产生过多的活性氧物种(ROS)。目前针对线粒体疾病仍缺乏有效的治疗方法。

复合体III的缺陷与高度可变的临床谱系有关。最近,研究人员表明,绿脓菌素,一种来自细菌的氧化还原酶,可以取代复合体III的氧化还原功能,起到电子分流的作用。亚微摩浓度的绿脓菌素是无害的,可以恢复呼吸,并被证明在5名患者的成纤维细胞中增加ATP的产生,这5名患者的TTC19、BCS1L或LYRM7存在致病性突变,这些蛋白参与了复合体III的组装/稳定。

此外,绿脓菌素使线粒体膜电位正常化,并轻度增加ROS的产生和生物生成。这些体外效应在果蝇TTC19KO和斑马鱼TTC19KD中都得到了证实,施用低浓度的绿脓菌素可以显著改善了机体运动能力

重要的是,每日给药,持续给药两个月的绿脓菌素对对照组小鼠没有毒性

因此,以上结果共同表明,利用氧化还原环素可以治疗复杂的线粒体复合物III突变相关的疾病。
