最近,有研究人员使用核心调控环路分析(CRC)鉴定了肾细胞癌(RCC)细胞中的一个候选致癌基因PAX8。大规模功能基因组验证筛选确定了PAX8的功能沉默能够导致RCC细胞系增殖减弱。PAX8依赖的顺反组表观遗传分析阐释了PAX8具有活性增强子元件,并控制各种代谢相关途径基因的表达。研究人员选择了铁氧化酶血浆铜蓝蛋白(CP)作为模范基因来解析PAX8的分子功能。研究发现,PAX8能够在CP启动子的增强子结合环路上招募组蛋白乙酰化活性。重要的是,CP的表达与PAX8沉默敏感性有关,并鉴定了具有不良生存的RCC案例亚群。

最后,研究人员指出,他们的数据鉴定了PAX8是RCC的一个候选致癌基因,并提供了控制活性的潜在生物标记。
