Nat Commun:结肠炎-->结肠癌转化过程中关键因子被发现

最近,我国研究人员发文证明,Fam3D的缺失与结肠粘膜的完整性受损、上皮增生增加、抗微生物肽的产生减少,以及对化学诱导的结肠炎的敏感性增加有关,并与癌症的高发病率有关。

炎症性肠病(IBD)是一种复杂的多因素疾病,包括克罗恩病和溃疡性结肠炎(UC),两者都以慢性复发性炎症和肠上皮损伤为特征,并与结直肠癌风险增加有关。UC是IBD的一种主要形式,目前,其病因仍不清楚。越来越多的证据表明,结肠粘膜屏障功能受损是导致UC发病的关键因素。

肠粘膜屏障的生理平衡是由遗传和环境因素共同维持的,其受损是炎症性肠病等疾病的发病机制之一。一种类似细胞因子的分子FAM3D(小鼠Fam3D)在小鼠胃肠道中高表达。最近,我国研究人员发文证明,Fam3D的缺失与结肠粘膜的完整性受损、上皮增生增加、抗微生物肽的产生减少,以及对化学诱导的结肠炎的敏感性增加有关,并与癌症的高发病率有关。

抗生素对Fam3D-/-小鼠进行预处理可显著降低化学诱导的结肠炎的严重程度,与Fam3D-/-小鼠共居的野生型(WT)小鼠表型为Fam3D-缺失,显示出对结肠炎的敏感性增加和粪便微生物群的构成偏斜。共居的WT和Fam3D-/-小鼠微生物群初始平衡,但在分离后细菌组成的分化越来越大。

这些结果表明Fam3D在结肠稳态、防止炎症相关癌症和正常微生物群组成中的重要作用。
