J Immunol:T胸腺中富集自身反应性B细胞

胸腺是T细胞分化的发生地,也是重要的淋巴器官。始祖T细胞从骨髓出来,经过皮质延髓束进入胸腺,并在其皮质区域进行TCR的重排。之后,表达TCR的这部分T细胞在胸腺中经历了两次重要的选择。首先,它们通过阳性选择检验了其表面的TCR与MHC的结合能力。其中,负责检验的是表面同时表达MHC-I与MHC-II的胸腺表皮细胞。之后,这部分通过了阳性选择的细胞再次经历阴性选择,这一步是为了检验T细胞与自身抗

胸腺表皮细胞表达的组织特异性自身抗原是由转录因子AIRE调控的,那些与自身抗原结合强度较高的T细胞将会诱发凋亡,从而保证免疫耐受的稳态。

虽然胸腺是T细胞分化的重要场所,但其中也存在大量的B淋巴细胞。它们在髓质区域聚集,并被T细胞所环绕。有研究表明这部分B细胞参与了自身抗原向T细胞的呈递。而且其表面表达MHC-II,能够有效呈递被BCR识别后的抗原物质。

B细胞主要在骨髓中分化完成,其中大部分B细胞都表达自身抗原特异性BCR。经过相似的选择过程,这部分细胞会被除去。最终离开骨髓进入循环系统的成熟B细胞中,20%存在自体反应性。

有研究表明,胸腺始祖细胞对于维持B细胞的潜能十分重要,也有文章指出胸腺中存在一定的B始祖细胞,而且这部分B细胞的BCR谱与骨髓来源的B细胞并不相同。

为了了解胸腺B细胞的细胞特性,来自荷兰鹿特丹医学中心大学的Menno C. van Zelm课题组进行了深入研究,相关结果发表在最近一期的《Journal of Immunology》杂志上。

首先,作者分离了小鼠胸腺的B细胞,并将其与骨髓来源的B细胞进行了比较。流式检测结果显示:小鼠胸腺中确实存在一定量的B细胞,而且这部分B细胞属于成熟但未激活的状态。

进一步,通过对B细胞内部BCR的重排过程进行分析,作者发现这部分细胞的BCR库与骨髓来源的B细胞并不相同。

之后,作者通过单细胞克隆技术分离了小鼠胸腺中以及骨髓中的B细胞,并将其与自身抗原进行反应。结果显示,胸腺中的B细胞大多具有自身抗原反应性。另外,作者发现这部分来自胸腺的B细胞具有多抗原反应性。
