Nature:百岁老人的肠道菌群如何异于常人?

最近,研究人员发现,百岁老人体内存在一个独特的肠道微生物群,富含能够产生独特的次级胆汁酸(BAs)的微生物。

微生物群通过其在控制消化功能、骨密度、神经元活动、免疫和抵抗病原体感染方面的作用,长期以来被认为是决定老年人健康状况的一个关键角色。老年人的微生物群往往显示出个体间的变异性增加和多样性减少,因此与免疫衰老、慢性全身性炎症和虚弱有关。

相比之下,百岁老人(100岁及以上)对与年龄有关的慢性疾病的敏感性较低,并可以在几次传染病的袭击中幸存下来。对此,有人推测,肠道微生物群中有一些百岁老人特有的成员,这些成员不仅仅是衰老的结果,还可能积极地促进对病原体感染和其他环境压力的抵抗。

最近,研究人员发现,百岁老人体内存在一个独特的肠道微生物群,富含能够产生独特的次级胆汁酸(BAs)的微生物,包括 iso-, 3-oxo-, allo-, 3-oxoallo-以及isoallo-利托胆酸(LCA)。

百岁老人的粪便中的isoLCA, 3-oxoLCA, alloLCA, 3-oxoalloLCA以及isoalloLCA明显升高

在这些BAs中,IsoalloLCA的生物合成途径以前未被描述。通过筛选一位百岁老人粪便微生物群中的68种细菌分离物,研究人员发现了Odoribacteraceae菌株在体外和体内都是isoalloLCA的有效生产者

此外,研究人员发现5α-还原酶(5AR)和3β-羟类固醇脱氢酶(3βHSDH)负责生产IsoalloLCA。IsoalloLCA可以对革兰氏阳性(但不是革兰氏阴性)多药耐药病原体,包括艰难梭菌和粪肠球菌,发挥了强大的抗菌作用

总之,这些发现表明,特定的胆汁酸代谢可能参与减少病原体感染的风险,从而可能有助于维持肠道的平衡。
