Blood:高γ'/总纤维蛋白原水平对VTE和缺血性卒中具有保护作用

纤维蛋白原是凝血级联反应的关键成分,其循环水平的变化可能导致血栓性疾病,如静脉血栓栓塞(VTE)和缺血性卒中。γ素(γ')纤维蛋白原是纤维蛋白原的一种异构体,具有抗凝特性。

Jillian等采用双样本孟德尔随机化(MR)方法,利用全基因组关联研究的汇总统计数据,评估总循环纤维蛋白原及其异构体γ'纤维蛋白原对VTE和缺血性卒中亚型风险的因果影响。选用γ'纤维蛋白原和总纤维蛋白原的遗传工具,在主分析中使用逆方差加权MR方法来估计因果效应,并辅之以对包括MR-Egger、加权中值MR和加权模式MR在内的多效性变体的更稳健的敏感性分析。

γ'/总纤维蛋白原水平与VTE和缺血性卒中的相关性

基于γ'纤维蛋白原的16种基因工具和总纤维蛋白原的75种基因工具的组合的主要逆方差加权MR评估表明,较高的γ'纤维蛋白原和总纤维蛋白原水平对VTE风险具有保护作用。此外,较高的γ'纤维蛋白原水平对心源性栓塞和大动脉卒中风险也具有保护作用。在敏感性分析中,因果效应的预估结果一致。

总之,该研究结果为基因决定的γ'纤维蛋白原在VTE和缺血性卒中风险中的作用提供了证据。未来需要进一步研究以探索这些效应的潜在机制及其临床应用。
