最近,有研究人员为了查找到这些因子,他们从颅骨中制备了条件培养基(CM)。通过活细胞成像技术,研究人员发现Calvarial-CM处理能够增加C4-2B4 PCa细胞的细胞静息。Calvarial-CM的质谱分析鉴定了132种分泌因子。蛋白免疫印迹和ELISA分析确认了一些因子的存在,包括了DKK3、BMP1、新生素和血管素。总颅骨与分离成骨细胞的qRT-PCR分析表明了DKK3、BMP1、新生素和血管素是主要是由成骨细胞表达,而MIA、LECT1、NGAL和PEDF则由其他颅骨细胞表达。重组的人类DKK3、BMP、血管素、新生素、MIA和NGAL处理能够增加C4-2b和C4-2B4 PCa细胞的细胞静息。同时,DKK3、血管素和新生素而非BMP1,能够通过激活p38MAPK信号途径来增加休眠。一致的是,DKK3、血管素和新生素不能诱导表达显性阴性p38αMAPK且BMP1仍旧有活性细胞的休眠,表明了BMP1通过另一个信号途径起作用。

最后,研究人员指出,骨分泌多重休眠诱导因子,这些因子能够通过远距离信号途径能够来诱导DTC休眠。
