干细胞因子SOX2、OCT4、KLF4和MYC诱导的信号途径不仅介导分化细胞多能性的重编程,同时与肿瘤恶性程度有关。

最近,有研究人员发现膀胱癌中,SOX2的表达预示者不良的无复发生存,并且与晚期病理等级相关。SOX2的沉默能够减弱膀胱癌细胞的生长,而其表达能够促进癌细胞的生存和增殖。在低血清应激下,SOX2的表达能够促进AKT的磷酸化和膀胱癌细胞的球体形成能力。另外,AKT磷酸化的药物抑制(MK2006)能够进一步的抑制SOX2介导的膀胱癌细胞球体的形成。基因表达分析表明SOX2的表达能够诱导IGF2的表达,而Sox2沉默则抑制IGF2的表达。更多的是,敲除IGF2和IGF1R能够减弱膀胱癌细胞的生长。最后,使用linsitinib抑制IGF1R同样也能够抑制低血清应激下SOX2介导的膀胱癌球体的形成。

最后,研究人员指出,他们的发现表明了SOX2-IGF2信号途径能够影响膀胱癌细胞生长的侵袭性。该信号途径可作为一个很有前景的生物标记和膀胱癌治疗靶标。
