Science:科学背书!从精液中发现的亚精胺,竟然有着抗衰老、抗癌、保护心血管和神经、改善肥胖和2型糖尿病等逆天神效

2018年1月,顶级期刊《科学》杂志为此发表了一篇重磅文章,综述了130多篇科研进展,详详细细地汇总了近些年来科学家们对于亚精胺神奇功能的研究,一句话概括下来就是,有关亚精胺的传说,竟然都有可能是真的。

1677年的一天,被誉为显微镜之父、微生物学开拓者的列文虎克(Antony van Leeuwenhoek),好奇地使用自制的显微镜观察自己的精液。

于是,理所当然地,列文虎克又多了一个“发现精子存在第一人”的头衔。在看到神奇小蝌蚪的兴奋之余,列文虎克还发现了精液中大量存在着一种结晶化合物。

1678年,列文虎克将这些发现报告给了英国皇家学会。1680年,凭借自制的显微镜、以及几乎看啥都是人类首次的优势,酿酒工人家庭出身、16岁即飘泊在外、后来找到一份“看大门儿”工作的列文虎克,正式成为英国皇家学会的会员。

然而,在之后的两百多年时间里,科学家们好像并不重视虎克在精液中的发现。直到1888年,德国化学家Ladenburg和Abel将列文虎克所提到的结晶化合物,命名为精胺(Spermine)。

1926年,这些结晶化合物的化学结构才被确定。除了精胺,科学家们发现精液中还存在着丰富的亚精胺(Spermidine)、腐胺和尸胺等多胺类物质,这也是形成精液特殊气味的原因。

当然,精胺、亚精胺也并非只存在于精液中。科学家们后来证实,在人体其它组织,甚至所有的真核细胞中,都广泛存在着精胺和亚精胺。还有一些常见的食物,比如大豆、蘑菇、奶酪等,也十分富含亚精胺。

戏剧性的是,在之后的几十年里,亚精胺从无人问津,一下子成为坊间广为流传的世间少有的补品。据说,食补亚精胺的神奇功效包括且不限于抗衰老、抗癌、保护心血管、改善高血压、增强免疫以及阻止老年痴呆……

2016年,诺贝尔生理学与医学奖授予阐明细胞自噬机制的日本科学家大隅良典。要知道在这之前,人们都是将自噬作为亚精胺如此神奇的主要理论支撑。这一下,让亚精胺在坊间更加瞩目,更加神奇。

按照正常的剧情,一个东西神到这种地步,应该也就离辟谣不远了。。。吧。

竟然不是辟谣,竟然还为亚精胺做了最权威的背书。

神秘的亚精胺

多胺,是指含有两个或更多氨基的化合物,几乎存在于所有的细胞中。腐胺、亚精胺和精胺都是参与多种已知和未知生物过程的多胺。今天故事的主角,亚精胺,就是由腐胺合成,并作为精胺的前体。

细胞内亚精胺的水平,取决于能从细胞外吸收多少、内源性合成多少,以及分解代谢多少。亚精胺的细胞外来源有两种,一是通过摄取食物获取,二是通过肠道微生物产生;亚精胺和精胺的内源性生物合成,则是需要鸟氨酸等前体物质,在相关酶的严格控制下逐步反应完成。

虽然科学家们目前并不清楚每一个细胞内都存在的多胺,究竟在整个生命活动过程中扮演着什么样的重要角色,但已经可以确定的一点就是,它们很重要且不可或缺。

1984年《衰老与发育机制》(Mechanisms of Ageing and Development)杂志上的一篇论文显示,科学家们注意到动物和人体器官、血液和尿液中的多胺水平,会随着年龄的增加而逐渐下降,尤其是亚精胺和腐胺。一些涉及多胺生物合成途径的酶,也显示出一致的趋势,在老年动物的组织中大大降低。

还有证据显示,外源性亚精胺的补充,可以延长包括酵母、线虫和果蝇等实验室模式生物的寿命,并且显着降低哺乳动物(小鼠)中与年龄相关的蛋白氧化损伤。有趣的是,当研究人员敲除细胞自噬必需基因时,发现亚精胺的寿命延长和抗衰老效应都消失了。这也就表明,亚精胺的部分功能,是通过自噬作用实现的。

所谓细胞自噬,可以简单理解为是细胞的维修系统。当一些细胞器出现衰老,或者一些蛋白结构受损出现功能障碍,那细胞就会通过囊泡,将废旧零部件送往溶酶体进行回收降解,并将原料再次利用。想想都觉得神奇,再想想如果细胞自噬过程出了障碍就更觉得可怕,要是细胞内的垃圾不能得到及时清理,不仅会加速衰老,更有可能导致细胞癌变、代谢异常等严重的疾病。

延缓衰老,延长寿命

如果亚精胺真的能够作为一款安全、高效的细胞自噬诱导剂,那封它一个长寿药的称号,也毫不为过。

2009年,《自然》子刊 Cell Biology 上一项研究证实,亚精胺诱导自噬基因相关转录物的显着上调,从而增强细胞自噬作用,抑制细胞氧化应激和坏死。并且补充亚精胺能够增加酵母、线虫和果蝇的寿命。 同年《实验老年医学》(Experimental Gerontology)杂志发表研究,指出富含多胺的饮食可以降低老年小鼠的死亡率。

2012年,British Journal of Pharmacology 上一项研究指出,口服补充天然多胺亚精胺和精胺,能够通过抑制抑制破骨细胞的形成(骨吸收),预防绝经后小鼠的骨质疏松。此外还有证据显示,亚精胺可以改善绝经期的下降。

2014年,Scientific Reports 研究表示,通过改善肠道菌群增加血液中亚精胺和精胺水平,可以防止小鼠身体和大脑老化,延长小鼠寿命。

2015年,Cell Metabolism 上发表了一项研究,发现多胺水平参与调节培养的细胞和小鼠的昼夜节律,并且补充亚精胺可以逆转小鼠随着年龄增长而出现的昼夜节律紊乱。除此之外也有研究表明,精胺、亚精胺增强干细胞功能,促进组织再生(促进毛发生长、延缓肌肉萎缩并改善肌肉再生)等。

当然,以上这些都是在动物模型中发现的亚精胺对于延缓衰老的证据,这些结论放在人类身上,是否也成立呢?



根据 Rejuvenation Research 期刊发表的一项针对意大利中部地区人群的横断研究显示,老年人全血中的亚精胺和精胺含量降低,但在百岁老人中,亚精胺和精胺仍然保持着和年轻个体一样的高水平。

虽然影响一位老人是否长寿的因素有很多,但起码现在看来,亚精胺和精胺就是其中一个。曾经有一项针对亚洲国家的流行病学研究也显示出,膳食补充多胺的总量与不同国家人类预期寿命长短显着相关。

血管保护

且不说正常情况下随着年龄的增长,心血管系统会越来越老化,现如今就算是年纪轻轻,也往往伴随着各种各样的心血管疾病。那具有抗衰老功效的亚精胺,在心血管领域有什么出色的表现呢?

2002年,《高血压研究》(Hypertension Research)上一篇文章指出,亚精胺补充剂能够减弱高盐饮食大鼠的高血压。2012年,《全球健康科学杂志》发表的一项涉及西方48个国家的大型流行病学研究,旨在研究膳食多胺对预防心血管疾病(CVD)的贡献。结果显示膳食多胺水平与心血管疾病死亡率之间呈负相关性。

2013年,Mechanisms of Ageing and Development 上一项研究指出,亚精胺能够通过提高NO的生物利用度、减少氧化应激和增强自噬,改善小鼠动脉内皮细胞的氧化损伤,并逆转小鼠的动脉老化。这也表明亚精胺的逆天功能也并非都是通过自噬作用实现的。

2016年,Atherosclerosis 发表研究证实,亚精胺还可以减少动脉粥样硬化斑块中的脂质积聚。同年,《自然医学》杂志发表一篇重磅研究,证实膳食亚精胺通过增强自噬发挥心脏保护作用,减少心脏肥大、改善舒张功能,并且能够延长小鼠寿命。研究人员还在人类中证实了这一结论,通过Bruneck队列研究人员证实,饮食中的亚精胺(或亚精胺和精胺结合)的摄入,能够改善血压,降低心血管疾病发病率、死亡率。

防癌、抗癌

20世纪70年代,有研究指出在皮肤癌、乳腺癌结肠癌肺癌和前列腺癌中可以发现多胺浓度的增加,因此许多研究人员将多胺认为是潜在的致癌物质。然而后续的一些研究又发现,多胺含量又与一些癌症风险呈负相关。

而且有足够的证据显示亚精胺的补充可以减少小鼠的肿瘤发生。比如改善肠道产生多胺的菌群,降低了老年雌性小鼠中可见皮肤肿瘤的发生率;饮食补充亚精胺降低了严重肝纤维化小鼠肝癌的发生率;补充精氨酸也能减慢小鼠结直肠肿瘤的生长。

有趣的是,研究人员在挖掘亚精胺与癌症之间的关系时,还发现了除增强细胞自噬外,亚精胺增强抗肿瘤免疫反应的证据。具体而言就是,亚精胺会消耗肿瘤床中的免疫抑制细胞(肿瘤中免疫抑制细胞的存在,会帮助肿瘤细胞在免疫监视下发生逃逸),比如Treg细胞,从而刺激和增强肿瘤发生时的免疫监视。

此外还有研究发现,介导亚精胺产生的亚精胺合成酶(SRM),似乎还是肿瘤相关巨噬细胞(TAM)在结直肠癌发生过程中抗肿瘤作用的决定因素。这些发现或许能够解释,为什么外源补充的亚精胺,在体内具有很好的防癌抗癌作用(可能是由免疫刺激作用介导的),而在体外实验中却出现了有利于癌细胞增殖的现象。

膳食补充多胺,包括亚精胺,究竟对肿瘤发生和发展有着怎样的影响,多大的影响,目前仍然是一个开放的问题。但不管现在的结论怎样,这都是一个值得深入的研究方向。

神经调节和神经保护

脊髓损伤的后果估计大家都清楚,当脊髓神经细胞的长轴突损伤后,大脑和身体之间的通讯被阻断,往往造成损伤部位以下的瘫痪。2009年,《神经科学杂志》发表文章指出,精氨酸酶I的上调和多胺合成的增加,在调理中枢神经系统损伤中起重要作用,研究还找到亚精胺促进体内神经再生的证据。

2011年,顶级专业期刊 Investigative Ophthalmology & Visual Science 一项研究发现,在自身免疫性脑脊髓炎(一种多发性硬化症的模型)的小鼠中,口服补充亚精胺可以通过抗氧化作用减缓疾病进展,并减少视网膜神经节细胞的损失,来改善视功能。此外,也有研究证明在小鼠体内视神经损伤后,亚精胺能够促进视网膜神经节细胞存活和视神经再生,并在正常眼压性青光眼的小鼠模型中减弱视网膜变性。

不仅如此,2013年《自然神经科学》(Nature Neuroscience)发现,简单的亚精胺喂养,不仅能够恢复老年果蝇体内的多胺水平,而且能够有效抑制年龄增长引起的记忆障碍。2014年,Cell Cycle 杂志发文证明,在无脊椎动物模型中,亚精胺可以挽救由α-突触核蛋白诱导的多巴胺能神经元损失,减轻帕金森病相关的神经变性。

总而言之,这些研究都提示了外源性亚精胺在多种神经退行性运动障碍和痴呆中潜在的神经调节和保护功能。至于底层机制,有可能是亚精胺通过作用于NMDA受体(N-甲基-D-天冬氨酸受体,是大脑中学习记忆中的关键物质)、自噬调节、炎症抑制以及某些未知过程实现的。

改善代谢性疾病

最后我们再来说说肥胖和2型糖尿病。近些年的一些研究也已经证明了多胺在改善代谢性疾病领域的巨大潜力。

2014年,《欧洲病理学杂志》一项研究揭示了多胺在能量平衡和葡萄糖代谢中的作用,补充精胺可以改善肥胖小鼠的葡萄糖利用率,并且能够显着降低体重。

2015年,《自然》杂志发表重磅研究,指出烟酰胺N-甲基转移酶的下调,激活鸟氨酸脱羧酶介导的亚精胺生物合成增加,导致小鼠白色脂肪组织的能量消耗增加,并且能够预防饮食诱导的肥胖症。同年,Genes & Development 上一篇研究也证明了这一点。

2017年,《自然》子刊《细胞死亡与疾病》也发现,亚精胺能够增强小鼠细胞自噬作用,减轻小鼠高热量饮食导致的体重增加,并改善肥胖相关的症状。

至于补充亚精胺是否可用于人类治疗肥胖和2型糖尿病,目前还不能下结论。但这已经成为未来研究的重要课题。

人类疾病诊断和治疗潜力

首先,可以明确的是多胺代谢途径的失调与多种类型的疾病密切相关。在阿尔兹海默病小鼠模型和轻度认知障碍的人类患者中,也观察到多胺和精氨酸代谢紊乱的证据,因此,这就提供了将多胺作为癌症、神经退行性疾病、中风、肾功能障碍,以及心力衰竭和心肌梗塞等疾病生物标志物的可能性,并以此作为疾病诊断和评估疾病演变的有力参考。

此外,动物实验中补充亚精胺能够改善健康和延长寿命的证据是足够的,而且一些人类流行病学结果也显示膳食亚精胺的相关益处。因此亚精胺等多胺,在一些慢性疾病、延缓衰老、健康改善等领域都具有巨大的治疗潜力。

随着世界人口的老龄化,慢性病糖尿病、心血管疾病、癌症和神经退行性疾病形势不容乐观。科学家们也在寻找一些强有力的干预措施,能够尽可能降低人口老龄化和慢性疾病蔓延对于人类社会造成的广泛影响。其中,饮食限制(CR)被认为是能够延缓衰老和改善健康的有效方案。

但饮食限制方案的局限性在于,限制强度难以把握,而且只有很少的人似乎能够长时间地改变他们的饮食习惯。因此,科学家们又开始寻找能够从药理学层面模拟饮食限制有益效果的卡路里限制性模拟物(CRMs),这样一种潜在可能的策略目前备受关注,也是科学研究的热门领域。

而这其中,精胺和亚精胺,就被认为是最强大的CRMs,因为它们不仅效果逆天,而且毒性极低。毕竟亚精胺是从细菌到人类的所有生物体中含有的丰富的天然多胺,因此,就算在小鼠中终生补充亚精胺没有任何副作用也就不足为奇了。

精胺和亚精胺外源性补充也特别简单,通过改变饮食增加富含亚精胺的天然食物,或者改善益生菌促进肠道中多胺的合成等,都可以有效增加人体亚精胺的水平。目前,德国柏林查理特大学的研究人员正在进行一项临床试验,以研究研究亚精胺补充对认知障碍患者认知功能影响的临床试验。

当然,着重声明一句,亚精胺补充与各种疾病改善的关系,并不一定是因果关系,所以在缺乏相关临床试验证据的情况下,还望谨慎。
