循环肿瘤细胞(CTCs)从原发肿瘤脱落到血液中,但其中只有一小部分细胞产生转移。

最近,研究人员在乳腺癌患者的CTCs中进行了一次体内基因组范围内的CRISPR激活筛选,以确定促进小鼠远处转移的基因。编码核糖体蛋白和翻译调节剂的基因在这次筛选中得到了富集。

编码大核糖体亚单位成分RPL15的过表达,增加了多个器官的转移性生长,并选择性地增强了其他核糖体蛋白和细胞周期调节剂的翻译。

对来自乳腺癌患者的新鲜分离的CTCs进行RNA测序,研究人员发现了一个具有强烈的核糖体和蛋白合成特征的子集;这些CTCs表达增殖和上皮标志物,并与不良的临床结局相关。

因此,靶向这种侵袭性的CTCs子集的治疗方法可能值得探讨,因为它是转移性进展的潜在抑制剂。
