Cell Metab:乳酸可以调节肌肉缺血再生

EC特异性的糖酵解调节剂pfkfb3可以缓解缺血性后肢再血管化和肌肉再生能力的受损。

内皮细胞(EC)产生的信号有助于器官再生,但已有的报导并没有描述血管内的分泌代谢沟通。

最近,研究人员发现,EC特异性的糖酵解调节剂pfkfb3可以缓解缺血性后肢再血管化和肌肉再生能力的受损。这是由于巨噬细胞采取原生血管性和原再生性M2样表型的能力降低引起的。

从机理上讲,pfkfb3的损失减少了ECs分泌的乳酸,降低了缺血肌肉中的乳酸水平。添加乳酸到pfkfb3缺陷的ECs中可以恢复MCT1依赖性的M2极化。

供应乳酸的ECs使巨噬细胞去促进肌肉祖细胞的增殖和融合。此外,乳酸盐极化的巨噬细胞的VEGF生产增加,导致正反馈循环,进一步刺激血管生成。

最后,在缺血过程中增加乳酸水平可以拯救巨噬细胞的分化,改善肌肉的再灌注和再生,而巨噬细胞特异性的mct1删除会阻止M2样分化。

综上所述,ECs利用糖酵解,为血管内分泌提供乳酸,引导肌肉缺血再生。
