Phytomedicine:地黄饮通过减轻外周炎症保护帕金森病模型多巴胺能神经元

验证中药配方地黄饮(DHY)对帕金森病(PD)小鼠模型的治疗作用,并探讨其潜在的分子机制。

背景:帕金森病(PD)是一种进行性神经退行性疾病,对人类健康的威胁越来越大,目前仍缺乏满意的治疗方法。最近,许多研究也证明了特定亚群的CD4+T细胞对PD病理学的影响。Th17细胞在PD的发病机制中起着重要作用,中医药已广泛应用于PD的临床治疗,在临床药物开发方面具有巨大潜力。

目的:旨在验证中药配方地黄饮(DHY)对PD小鼠模型的治疗作用,并探讨其潜在的分子机制。

方法:通过行为学和组织病理学测试验证DHY对1-甲基-4-苯基-1,2,3,6-四氢吡啶(MPTP)诱导的PD小鼠模型的影响。高分辨率质谱结合分子网络技术应用于DHY的物质分析。基于DHY的化学成分,进行了网络药理学研究。免疫荧光法和ELISA法检测细胞因子在外周免疫系统的表达。qPCR和免疫荧光法检测中枢神经系统炎症浸润

结果:DHY在MPTP诱导的PD小鼠模型中改善运动功能防止多巴胺能神经元的丢失。基于MS/MS数据和分子网络,鉴定或初步表征了DHY的118种成分。网络药理学认为IL-17信号通路神经活性配体-受体相互作用是重要的靶点。

与MPTP中毒小鼠相比,DHY组脾细胞Th17细胞数量减少,血清中IL-17A水平降低。另一方面,DHY治疗小鼠中脑的炎症浸润较少,这可能与周围炎症减轻有关。

结论:虽然DHY的潜在药理机制仍然缺乏,但该研究提供了证据表明DHY煎剂可以通过减轻外周炎症来保护多巴胺能神经元
