抗修饰蛋白抗体(AMPA)结合的表位库包括修饰的IgG表位,IgG被定位为连接RF和AMPA的不同反应性的共同抗原。

目的类风湿性关节炎(RA)患者通常会产生类风湿因子(RFs)、结合IgG Fc的抗体和抗修饰蛋白抗体(AMPA)AMPA是一种多反应性自身抗体,通常与瓜氨酸化、高瓜氨酸化和乙酰化抗原结合。最近,在RA中发现了结合含瓜氨酸的IgG表位的抗体,这表明可能存在其他未发现的IgG表位,并且IgG可能是RFAMPA的共有抗原。本研究旨在揭示风湿性疾病中的新IgG表位,并确定多反应性AMPA是否与IgG结合。

方法:使用来自RA、系统性红斑狼疮、干燥病(SjD)或脊柱关节病患者的血清,通过肽阵列评估IgG与天然、含瓜氨酸和含高瓜氨酸的IgG恒定区线性表位的结合,具有高度结合的表位通过酶联免疫吸附试验(ELISA)进一步评估。还通过ELISA评估单克隆AMPAIgG衍生肽和IgG Fc的结合。
