Nat Commun：国内科学家发现广谱新冠病毒中和抗体

8月17日，由中国科学院微生物所严景华研究员团队、华中科技大学生命与科学技术学院王晨辉教授团队，中国食品药品检定研究院王佑春教授团队，中国疾控中心病毒病所谭文杰教授团队合作研发出一人源化基因工程单抗，该抗体靶向人血管紧张素转化酶 2 (hACE2)受体，细胞和动物试验结果表明可显著中和SARS-CoV-2病毒以及同属SARS-CoV病毒，具有广谱中和特点。该研究成果对于新冠肺炎病毒的抗体治疗，尤其针对目前多种变异株具有重大临床应用价值。相关成果发表在国际期刊Nature Communications。

新型严重急性呼吸系统综合症冠状病毒 2 (SARS-CoV-2) 变异株不断出现，且传播速度愈加增强，如此的变异和传播速度对该病毒的预防控制提出挑战，亟需可以应对万变的有效措施进行治疗。中和抗体疗法已被证明有效，但变异株的出现，则单一位点的单抗必然失效，广谱中和抗体的研发则提出日程。
幸运的是，近日我国科学家已经成果分离出一株人源化的基因工程单抗（ h11B11 ），该单抗针对人血管紧张素转化酶 2 (ACE2) 受体，作为新冠肺炎病毒入侵宿主的受体的拮抗剂，该抗体表现出优越的广谱的、中和活性。可应对目前流行的各种变异株。

在 hACE2 小鼠模型中进行治疗或预防性给药时，h11B11抗体可减轻和预防 SARS-CoV-2 复制和病毒引起的病理综合征。在食蟹猴中注射多次高剂量的 h11B11 后，未观察到血压和血液化学毒理学的显着变化。h11B11/ACE2 和受体结合域 (RBD)/ACE2 复合物的结构分析显示 MAb 和 RBD 对受体的阻碍和表位竞争。
