尽管近年来,肺腺癌(LUAD)的基因组研究取得了显著进展,但仍有很大比例的LUAD患者仍缺乏针对性的治疗策略。而由于蛋白质是生命活动的直接执行者,因此对LUAD进行全面的蛋白质组学研究,可以填补基因组异常与致癌蛋白质功能之间的知识空白。近日,中科院等国内多家机构的科学家首次在国际上揭示了肺腺癌蛋白质组完整图谱,为肺腺癌的精准医疗提供了宝贵资源。

这项研究通过蛋白质组学,磷酸化蛋白质组学,转录组和全外显子组测序数据的综合分析,揭示了LUAD相关分子特征、其与临床结果的关联、潜在的预后生物标志物和药物靶标,并于7月9日在线发布在《Cell》上。题目为“Integrative Proteomic Characterization of Human Lung Adenocarcinoma”

为了全面了解LUAD的分子结构,并为LUAD更精确的诊断和治疗提供基础,科学家们在本研究中对103例LUAD患者的肿瘤组织及其对应的非癌性邻近组织进行了蛋白质组学和磷酸蛋白组学分析。共鉴定出11119个蛋白质和22564个磷酸化位点。将这些蛋白质组学数据与临床信息和基因组学数据相结合,得到了LUAD的一个全面的分子图谱。

科学家们发现,上皮-间质转化以及炎症和致癌信号通路,很可能导致I期LUAD的不良预后。他们进一步建立了EGFR或TP53突变患者的基因改变和蛋白质组学特征之间的联系,EGFR或TP53是中国LUAD患者的两个主要驱动基因。

在EGFR突变患者中,观察到两种组织学标志物TTF-1和Napsin-A以及剪接体中蛋白质的表达水平升高。在TP53突变患者中,包括DNA复制和错配修复在内的多种致癌途径发生了显著变化。

研究人员将所有患者分为三种蛋白质组亚型(S-I,S-II,S-III),每种亚型均具有独特的分子特征和临床特征。全面的生物信息学分析显示,S-I患者与细胞环境和良好的预后相关。相比之下,S-III患者与细胞增殖和蛋白质组稳定性以及预后不良有关。磷酸蛋白质组学分析进一步揭示了不同蛋白质组学亚型中激活的信号通路和激酶。蛋白质组学数据和临床数据的综合分析进一步揭示了27种潜在的血浆生物标志物和LUAD的多个药物靶标。其中,研究人员指出,HSP 90β是潜在的生物标志物之一,并已在一个独立队列中得到进一步验证。

这项工作为LUAD的研究提供了新见解,超越了当前的基因组研究。中国科学家在国际上首次对肺腺癌开展了大规模、高通量、系统性的全景蛋白质组学研究,在基因组学,转录组学,蛋白质组学和磷酸化蛋白质组学水平上建立了中国LUAD的完整图谱。该蛋白质组数据集作为一种宝贵和强大的资源,将加快基础研究转化为临床实践中更精准的诊断和治疗。
