JCC:间充质干细胞疗法可诱发先天性免疫记忆反应从而减轻结肠炎

背景和目标:间充质干细胞(MSCs)由于其免疫调节特性而被用于治疗免疫介导的疾病。MSCs的细胞疗法在免疫系统中诱导多种作用,最终导致具有调节表型的免疫细胞数量增加。在这项研究中,研究人员调查了在临床相关的结肠炎小鼠模型中,MSC的有益效果是否能长期维持。

方法: 在疾病的诱导阶段,将单剂量的脂肪来源的MSC(aMSC)注入到葡聚糖硫酸钠(DSS)诱导的大肠杆菌小鼠中。在十二周的潜伏期后,以第二个7天的DSS周期再次评估小鼠的结肠炎。

结果: 与未用aMSC处理的DSS诱导的结肠炎小鼠相比,用aMSCs处理的DSS诱导的结肠炎小鼠在第二次DSS诱导期间表现出明显的肠道疾病活性指数降低。令人惊讶的是,在没有发生适应性免疫记忆细胞应答的Rag-1 -/-小鼠中也观察到了由aMSC疗法诱导的长期保护。首次炎性攻击后12周,在注入aMSC的DSS诱导的结肠炎小鼠的腹膜腔,脾脏和骨髓中,发现Ly6G + CD11b +髓样细胞的百分比增加。有趣的是,在用DSS再次攻击后,这些动物在结肠固有层中的调节/炎症巨噬细胞比例随之增加。

结论 本现研究结果首次证明,MSC治疗可以在小鼠中产生先天性免疫记忆样反应,从而长期提供对急性炎症的持续保护。
