CLIN CANCER RES近期发表了一篇文章，报道了靶向ENPP3的抗体共轭于MMAF形成的抗体-药物共轭物（ADC）治疗晚期转移性肾细胞癌的安全性，药代动力学以及推荐Ⅱ期实验剂量。

CLIN CANCER RES近期发表了一篇文章，报道了靶向ENPP3的抗体共轭于MMAF形成的抗体-药物共轭物（ADC）治疗晚期转移性肾细胞癌的安全性，药代动力学以及推荐Ⅱ期实验剂量。

作者使用了两种ADC进行了两个Ⅰ期临床试验，两个药物分别为AGS-16M8F和AGS-16C3F。AGS-16M8F在0.6-4.8mg/kg范围内的5个剂量水平进行静脉输入直至出现疾病进展或不可耐受毒性。这一队列在达到最大耐受剂量终止。AGS-16C3F在4.8mg/kg剂量开始进行试验。研究结果表明，AGS-16M8F队列（n=26）中位治疗持续时间为12周。1例患者出现持续的部分缓解，1例患者出现病情稳定时间延长。在AGS-16C3F队列（n=34）中，方案确定的最大耐受剂量为3.6mg/kg，但是无法耐受。可逆性的角膜病变限制了剂量。1.8mg/kg剂量最后被认为是安全的且均有抗肿瘤活性。接受1.8mg/kg剂量治疗的13例患者中有3例出现持续的部分缓解。2.7mg/kg以及1.8mg/kg的8例患者出现疾病控制＞37周。

文章最后认为，AGS-16C3F在1.8mg/kg时可耐受且具有持续的抗肿瘤活性。
