三氯生(TCS)作为一种防腐和消毒剂,通过大量的消费品和不断增加的环境释放量与人类直接接触。研究人员已经证明,TCS会促进小鼠肝脏肿瘤的发生,但至今为止,TCS发挥其毒性的生物和分子机制,特别是在肝病的早期阶段,在很大程度上还没有被研究出来。

最近,研究人员在PNAS杂志发文,其发现,当给小鼠喂食高脂饮食(HFD)时,脂肪肝和血脂异常是TCS诱发肝脏异常的突出早期症状。据推测,HFD诱导的代谢调节因子成纤维细胞生长因子21(FGF21)的肝脏表达被TCS钝化。TCS改变的Fgf21表达与编码代谢酶的基因的异常表达一致,表现为深刻的全身代谢变化,扰乱氨基酸、脂肪酸和葡萄糖的平衡。

在1型糖尿病动物模型中,TCS能增强和加速脂肪性肝炎和纤维化的发展,同时伴有肝脏脂滴和氧化应激水平的增加。粪便样本分析显示,HFD喂养的小鼠粪便物种丰富度降低,TCS进一步降低了微生物多样性,并使细菌群落向较低的Bacteriodetes和较高的Firmicutes转变,类似于非酒精性脂肪性肝炎(NASH)患者微生物群落组成的变化。
使用反向遗传学方法,研究人员证明,伴随着HFD,TCS诱导肝脂肪变性和脂肪性肝炎共同受转录因子ATF4和核受体PPARα调控,它们参与Fgf21基因的转录调控。
因此,该研究提供了营养失衡和TCS暴露与NASH进展相关的证据。
