近日,美国弗吉尼亚大学的研究人员在《Genome Research》上发表了一项新研究,揭示了男性和女性体内脂肪的遗传差异如何影响男女可能罹患的疾病。

具体而言,研究人员分析了大约3000个不同地理和种族的人类样本,研究皮下脂肪组织转录调控中的性别差异,最终鉴定出了162个在表达中存在强烈性别差异的基因,这些基因与脂肪存储和脂肪形成有关。

其中,有92个基因在肥胖者的脂肪组织中差异表达;24个基因在内脏脂肪组织中差异表达;70个基因在至少一种代谢活跃的组织中差异表达(包括内脏脂肪、肝脏、骨骼肌、左心室、心房、血液和13个脑组织)。

此外,在男性中表达水平升高的差异表达基因与包括雄激素和雌激素受体(AR和ESR1)在内的性激素受体相关,而女性中表达升高的差异基因与成脂转录因子 CEBPB和PPARG显着相关。

经过进一步研究,研究人员发现了6个特定基因,这些基因在调节脂肪组织的活性方面有特别的影响,分别为FADS1、MAP1B、HSPA12A、CLIC6、MMD和PDZD2。因此,可以将这6个基因作为潜在的治疗靶点。

作者强调,脂肪组织中的基因表达会影响脂肪的分布方式,而这种分布方式会影响2型糖尿病血管疾病的风险。男性的有害脂肪分布模式与心脏代谢疾病风险相关,而女性的脂肪分布模式则使得肥胖患病率较高。

尽管许多研究都表明了脂肪细胞信号传导和生理机制存在性别差异,但人类脂肪组织中基因表达的遗传基础是否存在性别差异还是未知数。因此,这项新发现从基因层面揭示了肥胖相关的性别差异,有助于解释男女面临的不同健康风险,并为更好、更有针对性的治疗奠定基础。
