Biomed Pharmacother:柴胡加龙骨牡蛎汤对伴焦虑症的急性心肌梗死的治疗作用

探讨柴胡加龙骨牡蛎汤(BFG)治疗急性心肌梗死伴焦虑的可能机制。

背景:焦虑血管疾病的常见共病,会恶化心脏功能。据报道,柴胡加龙骨牡蛎汤(BFG)具有抗氧化性能,可减轻心肌缺血损伤,改善焦虑样行为。核因子红细胞2相关因子2(Nrf2)/血红素加氧酶-1(HO-1)通路是防御氧化应激和改善心脏功能的主要机制。该研究旨在探讨BFG治疗心理-心脏病的可能机制。

方法:结扎左冠状动脉前降支,结合不确定性空瓶饮水刺激法,建立AMI伴焦虑大鼠模型,给予BFG(1 mL/100 g/d灌胃)或富马酸二甲酯(DMF,10 mg/kg/d腹腔注射)6天(DMF是一种Nrf2激活剂)

超声心动图、心肌损伤标记物、H&E和Masson染色用于评估心脏功能。行为测试和海马神经递质用于记录焦虑样行为。采用免疫组织化学、RT-PCR、western blotting和生化分析来检测Nrf2/HO-1通路相关因子的蛋白和基因表达,以及氧化应激凋亡参数。

结果:AMI组和复合组大鼠出现心脏功能恶化,以及焦虑样行为。BFG改善了超声心动图指标,降低了心肌损伤标记物,并减轻了心肌病变。BFG还可以改善焦虑样行为,提高神经递质水平。BFG促进了Nrf2/HO-1通路的激活,提高了抗氧化酶活性,降低了脂质过氧化水平,并减轻了氧化损伤和细胞凋亡。DMF具有与BFG相似的治疗作用和分子机制。

结论:BFG可能通过激活Nrf2/HO-1通路,抑制氧化应激和细胞凋亡,对伴有焦虑症的AMI患者具有心理-心脏病学治疗作用
