基因改造的自然杀伤(NK)细胞治疗胶质母细胞瘤

胶质母细胞瘤是成人中最常见的恶性脑肿瘤,目前的治疗选择极为有限且通常无效,导致大多数患者在确诊后12-18个月内死亡。近期,美国普渡大学的科研人员已经开发出一种有前途的新方法,即使用自然杀伤(NK)细胞来治疗胶质母细胞瘤,研究人员对NK细胞进行了基因修饰,使其能够精确靶向并杀死癌细胞。

普渡大学医学院的助理教授Sandro Matosevic表示:“对NK细胞进行多功能改造是改善治疗方法的潜在方式”。开发针对GBM(多形胶质母细胞瘤)的疗法所面临的挑战是该疾病具有高度免疫抑制作用,GBM细胞使用多种机制来逃避免疫系统的识别,因而针对它开发的许多疗法均未取得成功。

GBM是最具侵略性的脑肿瘤。尽管有多种强化疗法(手术、化疗和放疗)可供选择,但GBM的患者很少能存活超过15个月,而且没有一例可以治愈。这是因为大多数药物通过单一途径起作用,而GBM使用高度复杂的免疫抑制机制来增强对治疗的抵抗力并逃避免疫识别。
