J Dent Res：间质SuFu通过Shh信号调节下颌磨牙的发育

研究通过Dermo1-Cre小鼠敲除牙间质中的Sufu导致下颌磨牙进入帽状期延迟并伴随早期釉结(EK)形成缺陷。间质Sufu突变后，牙上皮中细胞增殖受到破坏并且发生程序性的细胞死亡。上皮-间质粘附分子E-cadherin在E14.5的牙胚双向基细胞中明显减少。EK中主要表达P-cadherin的细胞出现分层但无法凝聚。并且，早期EK标志基因的转录与对照组相比明显减少，包括：Shh， Fgf4和p21。相反，Sufu缺陷导致间质内Shh信号的上调，表明Gli1和Ptch1的显著上调。同时，间质-上皮诱导的关键因子Bmp4和Fgf3在牙间质中受到明显抑制。此外，Runx2在蕾状期的表达出现了瞬时减少。

总的来说，这些结果表明，间质Sufu对于Shh信号起到重要的调控作用，它可能是Bmp4和Fgf3信号的上游调节因子，协调牙胚间质和上皮之间的相互作用。
