GUT: 单核细胞通过α4β7整合素归巢后介导巨噬细胞依赖性肠伤口愈合

设计
研究人员通过流式细胞仪和免疫组织化学研究了IBD患者外周血单核细胞亚群和肠道中巨噬细胞亚群中归巢标记的表达。整合素功能通过动态粘附测定和体内肠道归巢测定来解决。在缺乏或缺失α4β7整联蛋白的小鼠中研究了体内伤口愈合。

结果
在归巢标记表达方面,经典和非经典单核细胞明显有差异,单核细胞表达的α4β7整联蛋白在功能上对粘膜血管地址动态粘附细胞黏附分子1和体内肠道归巢功能具有重要作用。受损的α4β7依赖性肠归巢受损与减少(效应大小约20%)和伤口愈合延迟以及伤口愈合巨噬细胞的病灶周围存在受到抑制有关。在接受维多珠单抗临床治疗的IBD患者中,外周血中非经典单核细胞增加。

结论
单核细胞的肠归巢功能受损可能导致伤口愈合巨噬细胞减少,并可能解释了接受维多珠单抗治疗的患者术后并发症发生率增加的现象。
