研究人员发现,p300/CBP相关的因子(PCAF)能够直接结合乙酰化δ-连环蛋白,然而一些I类和II类HDACs能够恢复上述影响。与β-连环蛋白不同,δ-连环蛋白能够通过PCAF调控的乙酰化下调,并通过HDAC调控的去乙酰化上调。HDAC抑制剂曲古菌素A能够减弱HDAC1调控的δ-连环蛋白的上调,而HAT或者自噬抑制剂而不是蛋白酶体抑制剂能够阻止PCAF调控的δ-连环蛋白的下调。这些结果表明了PCAF调控的δ-连环蛋白乙酰化能够促进其以Atg5/LC3依赖的方式自噬降解。缺失和点突变鉴定了一些δ-连环蛋白结构域中的赖氨酸残基参与了PCAF调控的δ-连环蛋白下调。PCAF在前列腺癌细胞中的过表达能够显著的减少δ-连环蛋白的水平和抑制细胞生长和运动。PCAF调控的δ-连环蛋白下调能够抑制E-钙粘蛋白加工和减少β-连环蛋白在细胞核内的分布,从而导致抑制β-连环蛋白/LEF-1调控的下游因子的表达。

最后,研究人员指出,这些数据阐释了PCAF能够通过促进δ-连环蛋白的细胞自噬降解下调δ-连环蛋白,并且能够抑制-连环蛋白调控的致瘤信号。
