急性髓系白血病(AML)是一种年龄相关性疾病,高度依赖于骨髓微环境。随着年龄的增长,组织中会积累衰老的细胞,特点是细胞增殖不可逆性阻滞和分泌一些列促炎性因子、趋化因子和生长因子,统称为衰老相关的分泌表型(SASP)。

Amina M Abdul-Aziz等人发现在骨髓微环境中,AML细胞可诱导基质细胞出现衰老表型,该表型是p16INK4a表达所驱动的。表达p16INK4a的衰老基质细胞又反过来通过SASP促进AML细胞存活增殖。重要的是,在活体内,选择性清除p16INK4a阳性的衰老骨髓基质细胞可提高白血病小鼠的存活期。

此外,研究人员还发现白血病驱动的衰老肿瘤微环境是由AML诱导的NOX2驱动的过氧化物所引起的。最后,研究人员通过p16-3MR小鼠模型发现,靶向NOX2可减少骨髓基质细胞衰老,进而减少AML增殖。

总而言之,本研究表明白血病产生的NOX2所驱动的过氧化物可促进骨髓基质细胞发生促肿瘤性的p16INK4a依赖的衰老表型。同时揭示了衰老微环境对白血病病理的重要意义。
