J Endod:降钙素基因相关肽对牙髓干细胞活力、增殖和分化的影响

牙髓炎是在近牙髓组织处或牙髓组织内细菌繁殖而导致的牙髓炎症反应。在晚期阶段,当炎症涉及到牙髓坏死,牙髓的保存则依赖于牙髓干细胞(DPSCs),它能够分化为成牙本质细胞样细胞并形成修复性牙本质。这篇研究中,作者评估了感觉神经元降钙素基因相关肽(CGRP)对DPSC活力和增殖以及DPSCs分化为矿化细胞能力的影响。

研究通过商业化手段获取DPSCs并施以不同浓度的CGRP,分别通过3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl tetrazolium bromide检测, trypan blue染色, 5-bromo-2'-deoxyuridine细胞增殖实验和caspase-3染色评估代谢活动、细胞活力、增殖和细胞凋亡。通过茜素红染色和成牙本质细胞标志物的定量mRNA表达评估DPSC的分化情况。

结果显示,CGRP以剂量依赖的模式减弱DPSC的代谢活动,它能够被CGRP受体拮抗剂CGRP 8-37所抑制。CGRP诱导的活力细胞比例的减弱与细胞增殖减弱有关,而与caspase-3依赖的凋亡无关。有趣的是,地塞米松诱导的DPSC向矿化细胞的分化既不受CGRP刺激而抑制也不受其影响而增加。

结论:神经肽CGRP对DPSC的增殖有抑制作用,但并不会增强或抑制DPSCs向矿化细胞的分化。结果表明,CGRP可能负向影响DPSC的功能以促成再生或组织修复进程。
